چکیده

In this article is presented a novel subunit model for the tropocollagen (TC) macromolecule, the monomeric unit of soluble collagen.1 The TC macromolecule is a rigid rod of definite length composed of three helical polypeptide strands that are equal (or very nearly equal) in length.2 4 The strands are of two types chemically, designated a1 and a2.4 Each macromolecule contains two al strands and one a2 strand.4 Our thesis is that each a1 strand is a repeating sequence of a subunit a,, and that the a2 strand is a repeating sequence of a shorter subunit 02. It is proposed specifically, after close examination of experimental data, that the subunits 01 and 02 have lengths in the ratio 7:5. This proposal leads to the "selflimiting" model for the TC macromolecule illustrated in Figure 1. In this model the strands terminate in register when each al strand comprises five a1 subunits and the a2 strand comprises seven 02 subunits. The over-all molecular length L is the "beat period" of the two subunit lengths,

Response of A Saline Solution Containing A Macromolecule To An External Electric Field

The dynamical behavior of a model for body fluids in response to an external electric field is computationally investigated for communication frequencies. The effect of an applied potential difference between two electrodes in a saline solution containing a rodlike macromolecule is studied by solving the Poisson and ion continuity equations simultaneously using the finite element method (FEM). ...

The Effects of Proteases on the Tropocollagen Macromolecule and on Its Aggregation Properties.

density distribution has recently been directly demonstrated in electron micrographs of formaldehyde-fixed SIS-type aggregates by Kuhn, K., U. Hofmann, and W. Grassmann (Naturwiss., 46, 512 1959). I For this purpose the segments were stained by exposure to 0.002 M uranyl nitrate for 1 minute and washed with distilled water before drying on the grid. Under these conditions the uranium in solutio...

Entropic elasticity controls nanomechanics of single tropocollagen molecules.

We report molecular modeling of stretching single molecules of tropocollagen, the building block of collagen fibrils and fibers that provide mechanical support in connective tissues. For small deformation, we observe a dominance of entropic elasticity. At larger deformation, we find a transition to energetic elasticity, which is characterized by first stretching and breaking of hydrogen bonds, ...

Atomistic and continuum modeling of mechanical properties of collagen: Elasticity, fracture, and self-assembly

We report studies of the mechanical properties of tropocollagen molecules under different types of mechanical loading including tension, compression, shear, and bending. Our modeling yields predictions of the fracture strength of single tropocollagen molecules and polypeptides, and also allows for quantification of the interactions between tropocollagen molecules. Atomistic modeling predicts a ...
